Please can anybody advise on the following queries for me ?....

If i have a lens with a dedicated Macro function and a 1:1 Magnification ratio - how can the magnification be increased ?...would using close-up tubes with the lens be a viable option ?.. accepting that there would inevitably be a slight loss of quality..

also (sorry),

can a DSLR be connected directly (by way of a adapter) to a Microscope ?.

Has anybody had any experience of such things or can advise ?

You can use extension tubes or a teleconverter to get closer/boost magnification. Any minor loss in quality is made up for by increased image size.

There are adapters for connecting cameras to microscopes.  I have never tried them.

You can also mount some lenses backwards to get amazing macro photos.  There are a few on this site.

Canon makes one macro lens that is 1X to 5X, does not focus and does not take any non-macro photos but the macros are amazing.
